چکیده

Increased immunization coverage, along with improved sanitization and safe water, is the most important public health intervention to improve child health in documented human history, resulting in a significant decrease in child mortality during the 20 century. Immunization is considered by the World Health Organization (WHO) as one of the most cost-effective of all health interventions, having the capacity to not only save but also to transform lives from the misery of infectious diseases. Since 1988, polio has been eradicated in 121 countries and is today endemic in only four countries. Despite the many achievements of vaccines in reducing the burden of disease, many infectious diseases remain endemic in large parts of the world, in particular, in developing countries as well as in many indigenous communities and areas of low socio-economic status within developed nations. There are two key aspects that need to be addressed for any vaccination to be successful: first, to increase the potential to generate a potent defence against diseases that evade the immune system, and secondly, to induce long lasting protective immunity following a single administration. Consequently, the development of vaccines has always been focused on effective induction of both humoral and cell-mediated immunity (CMI). Most of the vaccines that are used today are delivered by intramuscular or

Recombinant probiotic-Based Vaccines; Attractive vehicles for immunization via mucosal routes

Introduction: The importance of genetically modified microorganisms, including probiotics, now widely have considered as attractive vehicles for mucosal delivery of therapeutic molecules. Previous studies have indicated that mucosal administration of these molecules can effectively trigger humoral along with cellular immune responses. Therefore, development of induction of mucosal immune respon...

Nasal Immunization by (PLGA) Nanospheres Encapsulated with Tetanus Toxoid and (CpG-ODN)

In induction of systemic and mucosal immunity, particulate antigens are more effective than soluble antigens possibly because they are more efficiently endocytosed by mucosal-associated lymphoid tissue (MALT) M cells. In this study, we determined the systemic and mucosal immune responses in rabbits following intranasal immunization of tetanus toxoid TT and CpG-ODN encapsulated within PLGA nanos...

Protective Immunity in Mice Following Immunization with the Cochleate-Based Subunit Influenza Vaccines

High morbidity and mortality of influenza virus infection makes it an important disease world-wide. Mouse is a very well-studied animal model for this disease with similar manifestation to human disease. It would be desirable to induce mucosal as well as circulating immune responses to obtain protection from infection and to decrease the spread of the virus. Cell mediated immunity (proliferativ...

Influence of mucosal and parenteral immunization with a replication-defective mutant of HSV-2 on immune responses and protection from genital challenge.

Herpes simplex virus (HSV) most frequently initiates infection at a mucosal surface; thus mucosal immune responses are likely to be important in defense against HSV infection. We have examined the effects of eliciting mucosal as well as systemic immune responses on protection against genital challenge infection with virulent HSV-2 in mice immunized with a replication-defective mutant of HSV-2. ...

Long-lived cytotoxic T lymphocyte memory in mucosal tissues after mucosal but not systemic immunization

The induction and maintenance of long-term CTL memory at mucosal surfaces may be a critical component of protection against mucosal pathogens and is one goal towards development of effective mucosal vaccines. In these studies we have functionally evaluated short and long-term CTL memory in systemic and respiratory or genital-associated lymphoid tissues following mucosal or systemic routes of im...
